The imported frozen storage tube is mainly composed of three parts: preservation liquid, preservation tube, and small porcelain beads. It is a container for laboratory preservation and is used for the preservation or transfer of bacteria. Generally, there are 25 small porcelain beads in the strain preservation tube for the adsorption and preservation of bacteria. During use, the bacteria were inoculated in a storage tube and placed in an a-20℃ or-80℃ environment for long-term storage. (-20℃ can save for one year, -80℃ can save for two years).
The procedure for imported frozen storage pipe:
1. Fresh cultures were picked from a bacterial pure culture with a suspension of about 3-4 mixers and inoculated in stock tubes.
2. Tighten the freezer tube, and reverse 4-5 times to emulsify bacteria.
3. Keep the freezer tube in the refrigerator (-20℃ - -70℃).
Suggestions for the safe use of imported frozen storage pipe:
1. When using the frozen storage tubes to store samples, it is strictly required to put the frozen storage tubes into the gas layer of liquid nitrogen or stored them in the refrigerator. If the frozen storage tube is stored in liquid nitrogen liquid, the liquid nitrogen has a certain chance that will penetrate into the frozen storage tube. During the recovery, the liquid nitrogen gasification leads to a pressure imbalance inside and outside the tube, which is easy to cause the burst of the frozen storage tube and has biological harmfulness.
2. Operate the frozen storage tube for resuscitation, and use safety protection equipment throughout the whole process. It is recommended to wear experimental clothes, and cotton gloves and operate on a safe test table. If conditions permit, wear protective goggles or a protective mask. Since the indoor temperature will be higher in summer than in winter, please take extra care.
3. During the storage process of frozen storage cells, the freezing temperature of the frozen storage tube should be uniform. Uneven freezing will lead to the production of ice plugs, which will inhibit the temperature transfer of the liquid on both sides, thus causing dangerous high pressure and damage to the reservoir tube.
4. The sample size of the reservation shall not exceed the maximum working volume required by the frozen tube.
